"Shotgun" IM-MS sequencing of HS oligosaccharides

An international team of researchers, led by Prof. Rebecca Miller from University of Copenhagen and Prof. Dr. Kevin Pagel from Freie Universität Berlin, developed a novel strategy to characterize complex glycosaminoglycans (GAGs) such as heparan sulfate (HS). The new methodology is based on “shotgun“ ion mobility-mass spectrometry sequencing (SIMMS2) which enables direct and unambiguous sequencing of HS oligosaccharides through assembly of overlapping fragments. SIMMS2 alleviates previous analytical limitations imposed by the isomeric nature of many HS oligosaccharides and was recently published in the journal Nature Communications. The study emphasizes the potential of SIMMS2 for the discovery of bioactive motifs with pharmacological qualities. The results might aid in the development of novel GAG-based drugs with improved properties for therapeutic application.
