By using small nano-ESI emitters, we were able to measure unfolding curves that are strikingly similar to those obtained with traditional solution-phase methods. Moreover, direct measurement using mass spectrometry enables picking out the folding intermediate, which is a challenge in ensemble-averaged techniques.
